PROJECT NARRATIVE Cannabis Use Disorder is a highly prevalent public health concern with limited treatment options, and the treatment options that are available are rendered less effective in the likely incidence of comorbid Major Depressive Disorder. The current proposal will provide insight into a shared biological mechanism that has been implicated in both Cannabis Use Disorder and Major Depressive Disorder which may be specifically involved in relapse. Results from this study can then be applied to future medications development for comorbid Major Depressive Disorder and Cannabis Use Disorder.
Project Narrative In the United States, millions of people use cannabis and there is now convincing evidence linking its use with depression and suicidal behavior. By combining human postmortem genomics, MPRA genetic engineering, and human induced pluripotent stem cell derived brain organoids, we propose to test the molecular and functional impact of cannabis use perturbing the genotype of MDD risk alleles. We hope to find insights into the mechanism of disease initiation and progression of MDD in people with substance use disorders.
